How to assign knob on minilab 3 to knob on a VST synth?

Is it possible to quickly assign a knob on the minilab 3 to a virtual knob on a virtual synth? For example the frequency knob on the Oberheheim Sem 3?

Yes. You have different possibilities.

There is a pre made MIDI config for MiniLab 3 in SEM V3. Just select MiniLab 3 as Midi Controller under the MIDI tab in the right panel. Open the panel using the GEAR ICON at the top right of the application.

One of the Macros might already be assigned to control the Cutoff.

I suggest you in SEM Manual read about MACRO’s (section “6.8. Macros” in the manual) - and MIDI configs section “7.3.2. MIDI Tab” and forward). You can create your own MIDI configs, if you wish.

EDIT: BTW. I forgot you posted in Analog Lab forum. You need to have the full version of SEM V3 to assign to SEM parameters through Analog Lab. In that case please read Analog Lab Manual section “23.5.3. Macros Tab”.
I think perhaps you shall use Macros. But try things out. I would try out the Brightness Macro. EDIT END.
